No. RKTX1MAG-37K) Toxicity is the leading cause of drug failure, so increasing the success rate of therapeutic development requires more sensitive and rapid methods to determine organ-specific damage. The traditional tests to determine kidney toxicity are blood urea nitrogen (BUN) and serum creatinine (SCr), which are not optimal for early detection or localization of kidney damage. As a result, researchers have turned to recently qualified nephrotoxicity biomarkers for more predictive toxicity analysis. We used these panels to efficiently profile response to gentamicin, a known nephrotoxicant in rats. By simply measuring biomarker concentrations in urine, we detected kidney toxicity (marked by increased Clusterin and KIM-1 levels) in as few as three days (Figure 3). Figure 3A and 3B. 3 groups of 5 adult male Sprague Dawley® rats, each injected intraperitoneally daily with either water, low-dose (70 mg/kg) gentamicin or high-dose (140 mg/kg) gentamicin. Day 0 urine collection is after the 1st dose, and subsequent collections are at days 3, 7 and 14. All KIM-1 nephrotoxicity biomarkers H2O were analyzed, but we low dose gentamicin show data here only for high dose gentamicin Clusterin and KIM-1.
